Yammy 3 cylinder 2 stroke (fumey)
Yamaha 3 cylinder 2 stroke running on 50: 1 oil mixture but is a little fumey. Yes it is a 2 stroke I know but the fumes upon starting is possibly not so good. Can the mixture screw be adjusted to help with eliminating some of the smokeness?

You haven’t said what hp motor or whether it is pre-mix or self oiling.
I always thought Yamaha pre-mix motors ran at 100:1 maybe check your manual.

3 cylinder 40 will be 50-1. Use genuine Yamaha oil if not doing so already.

Yep
brother's 50 hp 3 potter the same thing , changed to yammie oil and it cleared up after about 6 trips. not sure why but i think the yammie oil and the merc premium oil seem to smoke less and idle smoother

if it's idling fine, don't adjust the mixture screws. that will only it make idle rough, or not at all!

I was once recommended to use Evinrude XD-100 i think it was don't hold it to me, for pre mix but it is pricey i personally use Valvoline i found it to have less smoke than Castrol
for some reason i love smelling 2 stroke motor bikes but 2 stroke outboards just make u choke and feel sick
